Presentation

The microscopy platform is in charge of managing the use, maintenance and fine-tuning of the microscopes. It also offers assistance in sample preparation, equipment management, data analysis, and experimental design.

Portfolio of services

Here below are indicated the imaging acquisition options available in our facility. These can be customized and others non-listed here can be studied and set-up under request. Please, contact the facility technician for further information.

  • Transmitted light imaging:
    • Image acquisition with bright field, phase contrast and differential interference contrast (DIC).
  • Fluorescence microscopy:
    • Image acquisition with filters for DAPI, GFP, YFP, Texas Red and Far Red.
    • Design and performance of multidimensional imaging experiments.
  • Confocal microscopy:
    • High resolution confocal fluorescence microscopy with tunable spectral detection (λ excit: 405, 458, 488, 514, 561 and 633 nm).
    • Super-resolution imaging with Airyscan module in the emission spectrum of the corresponding excitation range wavelength (λ excit: 405, 458, 488, 514, 561 and 633 nm).
  • Live-cell imaging:
    • Temperature, CO2 and humidity-controlled incubation chamber for widefield fluorescence.
    • High-sensitivity dual CCD cameras (Zeiss Axiocam 503 mono and AxioCam MRc).
    • Time lapse and long-term time lapse (eg multiple days) experiments for cultured cells with simultaneous transmitted and fluorescence illumination.
  • High-throughput microscopy:
    • Randomized acquisition capability in predefined cell culture formats.
    • Motorized stage for simultaneous multi-position time lapse recording.
  • Image processing, analysis and quantification:
    • Measurement of object intensity, size, number etc.
    • 3D reconstuctions, rendering and measurements.

Resources

The Platform has two inverted microscopes equipped with a fluorescence module.

Two microscopes have an incubation chamber with control of the percentage of CO 2 , humidity and temperature to provide the necessary environmental conditions for conducting multidimensional in vivo "time-lapse" experiments with cell cultures.

The platform offers the possibility of acquiring high resolution images using structured lighting systems with the Zeiss ApoTome.2 module and of achieving super-resolution values ​​thanks to its Zeiss Airyscan module.

Zeiss LSM 880 Airyscan

  • Inverted microscope Axio Observer.Z1
  • Plan-Apocromat objectives 5x, 10x, 20x, 40x Oil, 63x Oil, 63x Water and 100x Oil.
  • Position control with motorized plate for horizontal movement and piezoelectric insert for vertical movement.
  • Zero-pixel-shift fluorescence lamp and filters for DAPI, GFP, Cy3 and Cy5.
  • AxioCam MRm camera.
  • LSM 880 scanning module.
  • Laser lines: 405, 458, 488, 514, 561 and 633 nm.
  • T-PMT transmitted light detector and optical filters for brightfield, phase contrast and interference differential contrast.
  • Three PMTs for fluorescence detection plus one Airyscan detector.

Zeiss Axio Observer.Z1 Apotome.2

  • Inverted microscope Axio Observer.Z1.
  • Objectives Plan; 5x, 10x, Neofluar 20x and 40x, and Apochromat 20x and 63x Oil.
  • Position control with motorized plate for horizontal movement and piezoelectric for vertical movement.
  • Optical filters for brightfield, phase contrast and interference differential contrast.
  • Zero-pixel-shift fluorescence lamp and filters for DAPI, GFP, Cy3 and Cy5.
  • AxioCam MRc and Axiocam 503 Mono cameras.
  • ApoTome.2 structured lighting module.
  • XLmulti S1 incubation cabinet.
  • XL S1 Incubation Cabinet Heating Unit, S1 Modules for CO 2 and temperature control.

Nikon Eclipse TE2000-E Microscope

  • Inverted microscope.
  • 10x, 20x, 40x, 60x and 100x objectives.
  • Transmitted light techniques (bright field, phase contrast and differential interference contrast) and fluorescence filters (DAPI, GFP, YPF and Texas Red).
  • Stage and revolving motorized objectives (X, Y, Z control).
  • Okolab incubation chamber with temperature, humidity and gas control.
  • Configuration with double digital camera for the acquisition of images with high resolution and fast capture.
  • Programmable automation of objectives, optical and fluorescence filters, sample positioning and auto-focus for "time-lapse" experiments.

Leica DMI 6000B Microscope

  • Inverted microscope.
  • 5x, 10x, 20x, 40x and 63x objectives.
  • Transmitted light techniques (bright field and phase contrast) and fluorescence filters (DAPI, GFP and Texas Red).
